Runes are consumable pickups with a wide variety of effects when used. They take up the same slot as pills, cards and objects when picked up. Their effects can be mimicked by using Clear Rune. For all intents and purposes, Soul Stones are considered runes too.
Fiend Folio adds 4 new runes to the game, 3 of which are Soul Stones.

Notes
- If Soul of Random would activate the effect of Soul of Lazarus, it will instead give that Soul Stone to the player to prevent unavoidable death.
- Ansus has a 1/1000000 chance of spawning in place of a regular Rune.
Unlock Conditions
- Soul of Random — Unlock half of all Soul Stones.
Trivia
- Soul of Golem's appearance is based off the item Golem's Orb.
- Soul of Random is based on the "random" option in the character select menu.
- The splash text "Who am I?" is identical to the text that appears over said menu.
- Ansus' name is a reference to the slang word "sus" that is synonymous with the game Among Us.
- This is detailed in Ansus being a sneaky "imposter" variant of Ansuz, intended to catch the player off guard.
- Ansus can be differentiated from a real Ansuz rune by its name, its description including punctuation at the end of "Vision", and the rune changing to point to the right while held in the pocket slot.
- There is a Soul of Slippy, Soul of Friend and Soul of the Fortune Teller, none of these can be spawned in-game even via the console.
